DNA vaccines would have numerous advantages in large animals and humans. In mice, they result in an endogen production of antigens and provide a balanced and effective immunity without requiring adjuvant, the production process is safe and simple and with quite a low cost, they can be used before birth, in utero, or immediately after birth without interference of passive immunity. This paper reviews all the progress made in the last decade in the field of DNA vaccines in terms of antigen expression and DNA vector penetration into the cell and the nucleus. The current major drawback of DNA vaccines is that, to date, none of the results obtained with mice could be reproduced in humans or large animals. However, there is too much at stake to give up what could be the third generation vaccines.
